knock a gene?

Question: Do scientist have the ability to knock a human gene yet, how to do it? is it simple?

Answer: If you mean to knock out (disable) genes, then it's tricky question. I'll tell you what happens when you knock out gene in other organism (plants, fungi, mouse, etc). You "build" an artificial DNA containing the gene of target. This gene is interrupted by other gene (a detector gene, with known functions which can be determined), in this way the gene of target can't be expressed and that other gene can be. The tricky part is that how you get this "construct" into human genome and the only possible answer for ethical and technical reasons is that you CAN'T.
